Q: Will the case interfere with s-pen function?

This isn't an answer. Does the case interfere with s-pen use on the screen. My current case makes the s-pen inoperative with the screen. If the case is removed s-pen works flawlessly Does this case interfere with s-pen?

Q: Is the back flat/flush or is the lock section raised creating a hump on the back of the case?

It is flat it is indented where it locks in

Q: Can anyone offer insight on how it compares to an otterbox defender in terms of drop protection? I hate switching cases all the time

Your choice to compare to the Otterbox case is clear. If you want a light and simple case, you have to buy an otter box case first, and if you want compatibility and convenience of quad lock, of course it is a quad lock case.
