Do you mix it with water and sugar or add it to fruit mixure straight away?

Hi You can do either but we normally recommend making a starter using a 1/3 of a cup of luke warm water and a couple of teaspoons of sugar stirrred in until it dissolves. Then add the yeast to this, cover it and leave it for 15 to 20 minutes to allow the yeast to start working. Then add this to the wine must. If you always make a starter then you know that the yeast was working when you added it to the must so that if anything goes wrong and the yeast dies / the wine does not ferment you know that it was not the yeast at fault and that it was a different factor such as too high or low a temperature, poorly sterilised equipment...... I have always used it at room temperature and it seems to work fine

would it be suitable for blackberry wine

Yes. I grow my own blackberries and make wine with them every year. Use the yeast for red wine.

What quantity of fruit/water??

You will need a recipe for your wine. Once you have that, you will see that the quantity of yeast is unimportant since it will multiply in the presence of sugar.
