Why does it take two capsules to deliver 1 mg of plasmalogen? Is 1mg even a clinically significant dose?

In a clinical trial (EBioMedicine 17:199-205(2017)), patients (aged 60 – 85 years) with mild AD were orally administered with either 0.5mg twice a day Scallop-derived PLASMALOGEN or placebo daily for 24 weeks and their cognitive function was assessed. Daily dosage of 1mg (0.5mg twice a day) plasmalogen was proven in another study (J Alzheimers Dis Parkinsonism. 9(4):474 (2019)) to be the optimal dosage to significantly improve body’s plasmalogen level and cognitive function (as assessed by MMSE test). Researchers found that higher dosages (5mg or 10mg daily) of plasmalogen did not result in significant improvement of cognitive function. It is highly recommended to take 0.5mg twice a day to get the optimal result.

What is the different between plasmalogen oil and plasmalogen extract?

Plasmalogen oil is made of plasmalogen precursor which must be converted into target plasmalogen molecules before being utilised by the body. Nonetheless, plasmalogen extract is already in the active plasmalogen form and hence, can be utilised by the body directly without further conversion. As a result, it is certainly that plasmalogen oil (plasmalogen precursor) will be required in much larger dosage than plasmalogen extract (active plasmalogen) as the conversion is required, which conversion efficiency is unknown and may vary between individuals.
